I am new to this program and it seems to be working great...taking small steps at the moment to figure everything out. My question is when I setup a new profile, how can I make the new profile load with my old settings - like friends lists, screen layout, etc. - or do I have to edit and save all this information all over again?

This is more of an EQ question than anything relating to WinEQ, but I can answer it anyway. EverQuest uses several "ini files" which store the friends list, UI layout, key settings, etc.
Your friends list, combat ability buttons and hotbuttons are stored in Character_server.ini in your EverQuest folder. Your UI settings are stored in UI_Character_server.ini. If you want to reuse the same settings for multiple characters, you need to copy these files from one character to another.
WinEQ profiles simply store information about the window size, what EQ folder to use, and what WinEQ hotkeys to use, and are completely separate from your actual EQ settings...

What layout and macros are you talking about? You're really confusing me now since I just explained that the UI and hotbuttons are EQ settings not WinEQ settings 
For each character in EQ, you have to set the layout of the game windows, your friends list, your hotbuttons, socials, etc. That has absolutely nothing to do with WinEQ
